This variant disrupts the p.Gly337 amino acid residue in COL1A2. Other variant(s) that disrupt this residue have been observed in individuals with COL1A2-related conditions (PMID: 27509835), which suggests that this may be a clinically significant amino acid residue For these reasons, this variant has been classified as Pathogenic. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt COL1A2 protein function. This variant has been observed in individual(s) with clinical features of autosomal dominant Ehlers-Danlos syndrome (Invitae). This variant is not present in population databases (ExAC no frequency). This sequence change replaces glycine with alanine at codon 337 of the COL1A2 protein (p.Gly337Ala). The glycine residue is highly conserved and there is a small physicochemical difference between glycine and alanine.
