Uncertain significance for DOCK2 deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004946.3(DOCK2):c.1873A>G (p.Met625Val), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces methionine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 625 of the DOCK2 protein (p.Met625Val). This variant is present in population databases (rs149106608, gnomAD 0.04%). This variant has not been reported in the literature in individuals affected with DOCK2-related conditions. ClinVar contains an entry for this variant (Variation ID: 1415380). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant¬†is likely to be tolerated.
