Uncertain significance for Glycogen storage disease type III — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000642.3(AGL):c.2033C>G (p.Thr678Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces threonine with serine at codon 678 of the AGL protein (p.Thr678Ser). The threonine residue is weakly conserved and there is a small physicochemical difference between threonine and serine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with AGL-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated.
