Uncertain significance for Left ventricular noncompaction 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001386795.1(DTNA):c.1529A>G (p.Asn510Ser), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the DTNA gene (transcript NM_001386795.1) at coding-DNA position 1529, where A is replaced by G; at the protein level this means replaces asparagine at residue 510 with serine — a missense variant. Submitter rationale: This sequence change replaces asparagine with serine at codon 480 of the DTNA protein (p.Asn480Ser). The asparagine residue is highly conserved and there is a small physicochemical difference between asparagine and serine. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. This variant has not been reported in the literature in individuals with DTNA-related conditions. This variant is not present in population databases (ExAC no frequency).
