Uncertain Significance for Hypertrophic cardiomyopathy — the classification assigned by All of Us Research Program, National Institutes of Health to NM_000258.3(MYL3):c.307G>A (p.Glu103Lys), citing ACMG Guidelines, 2015. This variant lies in the MYL3 gene (transcript NM_000258.3) at coding-DNA position 307, where G is replaced by A; at the protein level this means replaces glutamic acid at residue 103 with lysine — a missense variant. Submitter rationale: This missense variant replaces glutamic acid with lysine at codon 103 of the MYL3 protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). Splice prediction tools suggest that this variant may disrupt RNA splicing. To our knowledge, functional studies have not been reported for this variant. To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with MYL3-related disorders in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.

This study involves interpretation of variants in research participants for the purpose of population health screening. Participant phenotype was not available at the time of variant classification. Additional details can be found in publication PMID: 35346344, PMCID: PMC8962531
