This variant lies in the NAGLU gene (transcript NM_000263.4) at coding-DNA position 1178, where G is replaced by A; at the protein level this means replaces arginine at residue 393 with histidine — a missense variant. Submitter rationale: This sequence change replaces arginine with histidine at codon 393 of the NAGLU protein (p.Arg393His). The arginine residue is weakly conserved and there is a small physicochemical difference between arginine and histidine. This variant is present in population databases (rs753409259, ExAC 0.009%). This variant has not been reported in the literature in individuals affected with NAGLU-related conditions.
