Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006231.4(POLE):c.6432C>G (p.Phe2144Leu), citing Ambry Variant Classification Scheme 2023: The p.F2144L variant (also known as c.6432C>G), located in coding exon 46 of the POLE gene, results from a C to G substitution at nucleotide position 6432. The phenylalanine at codon 2144 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
