NM_001365951.3(KIF1B):c.391A>G (p.Asn131Asp) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.N131D variant (also known as c.391A>G), located in coding exon 4 of the KIF1B gene, results from an A to G substitution at nucleotide position 391. The asparagine at codon 131 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. The evidence for this gene-disease relationship is limited; therefore, the clinical significance of this alteration is unclear.
