Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006231.4(POLE):c.910-14A>G, citing Invitae Variant Classification Sherloc (09022015): This sequence change falls in intron 9 of the POLE gene. It does not directly change the encoded amino acid sequence of the POLE protein. RNA analysis indicates that this variant induces altered splicing and may result in an absent or altered protein product. This variant is present in population databases (no rsID available, gnomAD 0.01%). This variant has not been reported in the literature in individuals affected with POLE-related conditions. ClinVar contains an entry for this variant (Variation ID: 1413864). Studies have shown that this variant results in activation of a cryptic splice site, and produces a non-functional protein and/or introduces a premature termination codon (internal data). For these reasons, this variant has been classified as Pathogenic.
