Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000548.5(TSC2):c.5273C>T (p.Ala1758Val), citing Ambry Variant Classification Scheme 2023: The p.A1758V variant (also known as c.5273C>T), located in coding exon 41 of the TSC2 gene, results from a C to T substitution at nucleotide position 5273. The alanine at codon 1758 is replaced by valine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
