Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_174916.3(UBR1):c.5092A>G (p.Thr1698Ala), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals with UBR1-related conditions. This variant is present in population databases (rs748150323, ExAC 0.002%). This sequence change replaces threonine with alanine at codon 1698 of the UBR1 protein (p.Thr1698Ala). The threonine residue is highly conserved and there is a small physicochemical difference between threonine and alanine. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
