Uncertain significance for Charcot-Marie-Tooth disease type 2R — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_015271.5(TRIM2):c.786A>C (p.Lys262Asn), citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. This variant has not been reported in the literature in individuals affected with TRIM2-related conditions. This variant is present in population databases (rs746187978, ExAC 0.002%). This sequence change replaces lysine with asparagine at codon 235 of the TRIM2 protein (p.Lys235Asn). The lysine residue is highly conserved and there is a moderate physicochemical difference between lysine and asparagine. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
