Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003042.4(SLC6A1):c.1635G>A (p.Met545Ile), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces methionine with isoleucine at codon 545 of the SLC6A1 protein (p.Met545Ile). The methionine residue is highly conserved and there is a small physicochemical difference between methionine and isoleucine. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt SLC6A1 protein function. This variant has not been reported in the literature in individuals affected with SLC6A1-related conditions. This variant is not present in population databases (ExAC no frequency).
