NM_001754.5(RUNX1):c.883T>C (p.Ser295Pro) was classified as Uncertain significance for Hereditary thrombocytopenia and hematological cancer predisposition syndrome associated with RUNX1 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces serine with proline at codon 295 of the RUNX1 protein (p.Ser295Pro). The serine residue is moderately conserved and there is a moderate physicochemical difference between serine and proline. This variant is present in population databases (rs780399121, ExAC 0.001%). This variant has not been reported in the literature in individuals affected with RUNX1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
