ClinVar contains an entry for this variant (Variation ID: 14115). This missense change has been observed in individual(s) with Laing early-onset distal myopathy (PMID: 15322983). In at least one individual the variant was observed to be de novo. This variant is not present in population databases (gnomAD no frequency). This sequence change replaces arginine, which is basic and polar, with proline, which is neutral and non-polar, at codon 1500 of the MYH7 protein (p.Arg1500Pro). For these reasons, this variant has been classified as Pathogenic. This variant disrupts the p.Arg1500 amino acid residue in MYH7. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 15556047, 18660445, 19412328, 24119082).
