NM_000264.5(PTCH1):c.832T>G (p.Trp278Gly) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.W278G variant (also known as c.832T>G), located in coding exon 6 of the PTCH1 gene, results from a T to G substitution at nucleotide position 832. The tryptophan at codon 278 is replaced by glycine, an amino acid with highly dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
