Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001023.4(RPS20):c.31G>A (p.Val11Met), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals with RPS20-related conditions. This variant is not present in population databases (ExAC no frequency). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0"). This sequence change replaces valine with methionine at codon 11 of the RPS20 protein (p.Val11Met). The valine residue is highly conserved and there is a small physicochemical difference between valine and methionine. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
