This variant lies in the GNPTAB gene (transcript NM_024312.5) at coding-DNA position 2990, where A is replaced by G; at the protein level this means replaces tyrosine at residue 997 with cysteine — a missense variant. Submitter rationale: This sequence change replaces tyrosine with cysteine at codon 997 of the GNPTAB protein (p.Tyr997Cys). The tyrosine residue is highly conserved and there is a large physicochemical difference between tyrosine and cysteine. This variant is not present in population databases (ExAC no frequency). This missense change has been observed in individual(s) with mucolipidosis II (PMID: 29872134).
