This variant lies in the BMPR1A gene (transcript NM_004329.3) at coding-DNA position 1333, where A is replaced by G; at the protein level this means replaces isoleucine at residue 445 with valine — a missense variant. Submitter rationale: This missense variant replaces isoleucine with valine at codon 445 of the BMPR1A protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). Splice site prediction tools suggest that this variant may not impact RNA splicing. To our knowledge, functional studies have not been reported for this variant. This variant has been observed in an individual affected with colorectal cancer (PMID: 28135145). This variant has been identified in 6/251494 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
