Uncertain significance for Peroxisome biogenesis disorder, complementation group 7 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002617.4(PEX10):c.908G>C (p.Ser303Thr), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the PEX10 gene (transcript NM_002617.4) at coding-DNA position 908, where G is replaced by C; at the protein level this means replaces serine at residue 303 with threonine — a missense variant. Submitter rationale: This sequence change replaces serine with threonine at codon 323 of the PEX10 protein (p.Ser323Thr). The serine residue is weakly conserved and there is a small physicochemical difference between serine and threonine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with PEX10-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The threonine amino acid residue is found in multiple mammalian species, suggesting that this missense change does not adversely affect protein function. These predictions have not been confirmed by published functional studies and their clinical significance is uncertain. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
