NM_002087.4(GRN):c.1355T>A (p.Val452Glu) was classified as Uncertain significance for Neuronal ceroid lipofuscinosis 11; GRN-related frontotemporal lobar degeneration with Tdp43 inclusions by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt GRN protein function. ClinVar contains an entry for this variant (Variation ID: 1410618). This variant has not been reported in the literature in individuals affected with GRN-related conditions. This variant is present in population databases (rs759134554, gnomAD 0.008%). This sequence change replaces valine, which is neutral and non-polar, with glutamic acid, which is acidic and polar, at codon 452 of the GRN protein (p.Val452Glu).
