NM_000219.6(KCNE1):c.287A>G (p.Gln96Arg) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023. This variant lies in the KCNE1 gene (transcript NM_000219.6) at coding-DNA position 287, where A is replaced by G; at the protein level this means replaces glutamine at residue 96 with arginine — a missense variant. Submitter rationale: The p.Q96R variant (also known as c.287A>G), located in coding exon 1 of the KCNE1 gene, results from an A to G substitution at nucleotide position 287. The glutamine at codon 96 is replaced by arginine, an amino acid with highly similar properties. This variant was reported in a sudden cardiac arrest ventricular fibrillation cohort, as seen in one individual with a normal QTc value (Leinonen JT et al. Int J Cardiol, 2018 Jan;250:139-145). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
