Uncertain significance for Hypertrophic cardiomyopathy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003803.4(MYOM1):c.4708+5_4708+8del, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the MYOM1 gene (transcript NM_003803.4) at 5 bases into the intron immediately after coding-DNA position 4708 through 8 bases into the intron immediately after coding-DNA position 4708, deleting this region. Submitter rationale: This sequence change falls in intron 36 of the MYOM1 gene. It does not directly change the encoded amino acid sequence of the MYOM1 protein. It affects a nucleotide within the consensus splice site. This variant is present in population databases (no rsID available, gnomAD 0.007%). This variant has not been reported in the literature in individuals affected with MYOM1-related conditions. Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing (PMID: 17576681, 9536098). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
