NM_001103.4(ACTN2):c.2411A>G (p.Asn804Ser) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.N804S variant (also known as c.2411A>G), located in coding exon 20 of the ACTN2 gene, results from an A to G substitution at nucleotide position 2411. The asparagine at codon 804 is replaced by serine, an amino acid with highly similar properties. This variant has been reported in an unexplained cardiac arrest cohort (Grondin S et al. Eur Heart J, 2022 Aug;43:3071-3081). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
