Uncertain significance — the classification assigned by Quest Diagnostics Nichols Institute San Juan Capistrano to NM_005732.4(RAD50):c.3716G>A (p.Arg1239Gln), citing Quest Diagnostics criteria: The RAD50 c.3716G>A (p.Arg1239Gln) variant has been reported in individuals with breast cancer (PMID: 26689913 (2015), 33471991 (2021), see also LOVD (http://databases.lovd.nl/shared)) and esophageal squamous cell carcinoma (PMID 24572942 (2021)). This variant has also been identified in reportedly unaffected individuals (PMID: 33471991 (2021), see also LOVD (http://databases.lovd.nl/shared)). The frequency of this variant in the general population (Genome Aggregation Database, http://gnomad.broadinstitute.org) is uninformative in the assessment of its pathogenicity. Analysis of this variant using bioinformatics tools for the prediction of the effect of amino acid changes on protein structure and function yielded conflicting predictions that this variant is benign or damaging. Based on the available information, we are unable to determine the clinical significance of this variant.
