Uncertain significance for Curry-Hall syndrome; Ellis-van Creveld syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_153717.3(EVC):c.1720C>G (p.Arg574Gly), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine, a(n) basic and polar amino acid, with glycine, a(n) neutral and non-polar amino acid, at codon 574 of the EVC protein (p.Arg574Gly). This variant is present in population databases (rs141820870, gnomAD 0.005%). This variant has not been reported in the literature in individuals affected with EVC-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
