Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001014987.2(LAT):c.323C>T (p.Ala108Val), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces alanine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 108 of the LAT protein (p.Ala108Val). This variant is present in population databases (rs781688554, gnomAD 0.01%). This variant has not been reported in the literature in individuals affected with LAT-related conditions. ClinVar contains an entry for this variant (Variation ID: 1409251). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
