Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt STK11 protein function. This sequence change replaces alanine with glycine at codon 398 of the STK11 protein (p.Ala398Gly). The alanine residue is weakly conserved and there is a small physicochemical difference between alanine and glycine. This variant is present in population databases (rs768058962, ExAC 0.04%). This variant has not been reported in the literature in individuals with STK11-related conditions.
