Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000719.7(CACNA1C):c.2429C>T (p.Thr810Met), citing Ambry Variant Classification Scheme 2023: The p.T810M variant (also known as c.2429C>T), located in coding exon 17 of the CACNA1C gene, results from a C to T substitution at nucleotide position 2429. The threonine at codon 810 is replaced by methionine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
