NM_182961.4(SYNE1):c.20234A>G (p.Lys6745Arg) was classified as Uncertain significance for Emery-Dreifuss muscular dystrophy 4, autosomal dominant; Autosomal recessive ataxia, Beauce type by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This variant is present in population databases (rs769256850, ExAC 0.02%). This sequence change replaces lysine with arginine at codon 6674 of the SYNE1 protein (p.Lys6674Arg). The lysine residue is highly conserved and there is a small physicochemical difference between lysine and arginine. This variant has not been reported in the literature in individuals with SYNE1-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C25").
