Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002899.5(RBP1):c.117del (p.Ser40fs), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Ser40Alafs*60) in the RBP1 gene. It is expected to result in an absent or disrupted protein product. However, the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in RBP1 cause disease. This variant is present in population databases (rs559625725, gnomAD 0.3%), and has an allele count higher than expected for a pathogenic variant. This variant has not been reported in the literature in individuals affected with RBP1-related conditions. ClinVar contains an entry for this variant (Variation ID: 1408379). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
