Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001287.6(CLCN7):c.1225C>T (p.Arg409Trp), citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This sequence change replaces arginine, which is basic and polar, with tryptophan, which is neutral and slightly polar, at codon 409 of the CLCN7 protein (p.Arg409Trp). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with autosomal dominant childhood onset osteopetrosis (PMID: 23302420). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt CLCN7 protein function.
