This variant lies in the PALB2 gene (transcript NM_024675.4) at coding-DNA position 2890, where G is replaced by A; at the protein level this means replaces glycine at residue 964 with arginine — a missense variant. Submitter rationale: This variant is denoted PALB2 c.2890G>A at the cDNA level, p.Gly964Arg (G964R) at the protein level, and results in the change of a Glycine to an Arginine (GGA>AGA). This variant has not, to our knowledge, been published in the literature as pathogenic or benign. PALB2 Gly964Arg was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, suggesting it is not a common benign variant in these populations. Since Glycine and Arginine differ in polarity, charge, size or other properties, this is considered a non-conservative amino acid substitution. PALB2 Gly964Arg occurs at a position that is conserved across species and is located in the region of interaction with POLH, RAD51, and BRCA2 and in the WD3 repeat region (UniProt). In silico analyses predict that this variant is probably damaging to protein structure and function. Based on currently available evidence, it is unclear whether PALB2 Gly964Arg is a pathogenic or benign variant. We consider it to be a variant of uncertain significance.
