This variant has been observed in individual(s) with ataxia-telangiectasia and breast cancer, melanoma, thyroid cancer and kidney cancer (PMID: 25122203, 26681312, 26845104). It has also been observed to segregate with disease in related individuals. This variant is not present in population databases (gnomAD no frequency). This variant results in the deletion of part of exon 17 (c.2553_2638+61del) of the ATM gene. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), and loss-of-function variants in ATM are known to be pathogenic (PMID: 23807571, 25614872).
