NM_000089.4(COL1A2):c.2500G>A (p.Val834Ile) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.V834I variant (also known as c.2500G>A), located in coding exon 40 of the COL1A2 gene, results from a G to A substitution at nucleotide position 2500. The valine at codon 834 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
