Uncertain significance for Distal myopathy with posterior leg and anterior hand involvement; Myofibrillar myopathy 5; Hypertrophic cardiomyopathy 26 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001458.5(FLNC):c.7186C>T (p.Pro2396Ser), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the FLNC gene (transcript NM_001458.5) at coding-DNA position 7186, where C is replaced by T; at the protein level this means replaces proline at residue 2396 with serine — a missense variant. Submitter rationale: In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. This variant has not been reported in the literature in individuals with FLNC-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change replaces proline with serine at codon 2396 of the FLNC protein (p.Pro2396Ser). The proline residue is highly conserved and there is a moderate physicochemical difference between proline and serine.
