Therefore, it has been classified as a Variant of Uncertain Significance. This variant disrupts the p.Thr34 amino acid residue in MPZ. Other variant(s) that disrupt this residue have been observed in individuals with MPZ-related conditions (PMID: 8797476, 31372974), which suggests that this may be a clinically significant amino acid residue. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. This variant has not been reported in the literature in individuals affected with MPZ-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change replaces threonine with serine at codon 34 of the MPZ protein (p.Thr34Ser). The threonine residue is highly conserved and there is a small physicochemical difference between threonine and serine.
