Uncertain significance for Fucosidosis — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000147.5(FUCA1):c.1178C>T (p.Ser393Leu), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces serine, which is neutral and polar, with leucine, which is neutral and non-polar, at codon 393 of the FUCA1 protein (p.Ser393Leu). This variant is present in population databases (rs149168482, gnomAD 0.007%). This variant has not been reported in the literature in individuals affected with FUCA1-related conditions. ClinVar contains an entry for this variant (Variation ID: 1405327). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt FUCA1 protein function with a negative predictive value of 95%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
