Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001734.5(C1S):c.1522G>A (p.Val508Met), citing Invitae Variant Classification Sherloc (09022015): This variant is not present in population databases (ExAC no frequency). This sequence change replaces valine with methionine at codon 508 of the C1S protein (p.Val508Met). The valine residue is highly conserved and there is a small physicochemical difference between valine and methionine. This variant has not been reported in the literature in individuals affected with C1S-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
