NM_006231.4(POLE):c.5472G>T (p.Trp1824Cys) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.W1824C variant (also known as c.5472G>T), located in coding exon 40 of the POLE gene, results from a G to T substitution at nucleotide position 5472. The tryptophan at codon 1824 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
