Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000548.5(TSC2):c.4272C>G (p.Asp1424Glu), citing Ambry Variant Classification Scheme 2023: The p.D1424E variant (also known as c.4272C>G), located in coding exon 33 of the TSC2 gene, results from a C to G substitution at nucleotide position 4272. The aspartic acid at codon 1424 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
