Pathogenic for Severe neonatal-onset encephalopathy with microcephaly — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001110792.2(MECP2):c.798_837del (p.Lys266fs), citing Invitae Variant Classification Sherloc (09022015): This variant disrupts the C-terminus of the MECP2 protein. Other variant(s) that disrupt this region (p.Gly269Alafs*20) have been determined to be pathogenic (PMID: 17089071, 16473305, 12111643, 10854091). This suggests that variants that disrupt this region of the protein are likely to be causative of disease. For these reasons, this variant has been classified as Pathogenic. This variant has not been reported in the literature in individuals with MECP2-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change creates a premature translational stop signal (p.Lys254Asnfs*22) in the MECP2 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 233 amino acid(s) of the MECP2 protein.
