This variant lies in the CLCN1 gene (transcript NM_000083.3) at coding-DNA position 618, where C is replaced by A; at the protein level this means converts the codon for tyrosine at residue 206 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: This variant is not present in population databases (ExAC no frequency). This variant has been observed in individual(s) with autosomal recessive myotonia congenita (PMID: 31566103). It has also been observed to segregate with disease in related individuals. For these reasons, this variant has been classified as Pathogenic. This sequence change creates a premature translational stop signal (p.Tyr206*) in the CLCN1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in CLCN1 are known to be pathogenic (PMID: 17932099, 22094069, 23739125).
