NC_000017.10:g.(?_15142778)_(15142938_?)del was classified as Pathogenic for Charcot-Marie-Tooth disease, type I by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exon(s) 4 of the PMP22 gene. This variant would be expected to be in-frame, preserving the integrity of the reading frame. A similar copy number variant has been observed in individuals with Charcot-Marie-Tooth disease (PMID: 26454100, 28382305). It has also been observed to segregate with disease in related individuals. Algorithms developed to predict the effect of variants on protein structure and function are not available or were not evaluated for this variant. Experimental studies have shown that a similar copy number variant affects PMP22 function (PMID: 28382305). For these reasons, this variant has been classified as Pathogenic.
