Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000018.10:g.57586871C>G, citing Invitae Variant Classification Sherloc (09022015): This variant is not present in population databases (gnomAD no frequency). This variant occurs in a non-coding region of the FECH gene. It does not change the encoded amino acid sequence of the FECH protein. This variant has been observed in individual(s) with erythropoietic protoporphyria (PMID: 15850836). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. It has also been observed to segregate with disease in related individuals. This variant is also known as -250G>C. Studies have shown that this variant alters FECH gene expression (PMID: 15850836). For these reasons, this variant has been classified as Pathogenic.
