Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_053025.4(MYLK):c.3855G>T (p.Lys1285Asn), citing Ambry Variant Classification Scheme 2023: The p.K1285N variant (also known as c.3855G>T), located in coding exon 20 of the MYLK gene, results from a G to T substitution at nucleotide position 3855. The lysine at codon 1285 is replaced by asparagine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
