Uncertain significance for Nephronophthisis — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001023570.4(IQCB1):c.1550A>G (p.Asn517Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces asparagine with serine at codon 517 of the IQCB1 protein (p.Asn517Ser). The asparagine residue is moderately conserved and there is a small physicochemical difference between asparagine and serine. This variant is present in population databases (rs774106373, ExAC 0.009%). This variant has not been reported in the literature in individuals affected with IQCB1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The serine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function.
