Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_032608.7(MYO18B):c.7544T>A (p.Ile2515Asn), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces isoleucine with asparagine at codon 2515 of the MYO18B protein (p.Ile2515Asn). The isoleucine residue is moderately conserved and there is a large physicochemical difference between isoleucine and asparagine. This variant is present in population databases (rs763657203, gnomAD 0.002%). This variant has not been reported in the literature in individuals affected with MYO18B-related conditions. ClinVar contains an entry for this variant (Variation ID: 1400419). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Not Available"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Not Available"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
