This variant lies in the RTEL1 gene (transcript NM_001283009.2) at coding-DNA position 3045, where G is replaced by C; at the protein level this means replaces glutamine at residue 1015 with histidine — a missense variant. Submitter rationale: This sequence change replaces glutamine with histidine at codon 1015 of the RTEL1 protein (p.Gln1015His). The glutamine residue is moderately conserved and there is a small physicochemical difference between glutamine and histidine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with RTEL1-related conditions.
