Uncertain significance for Deficiency of adenosine deaminase 2 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001282225.2(ADA2):c.1307T>C (p.Met436Thr), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces methionine, which is neutral and non-polar, with threonine, which is neutral and polar, at codon 436 of the ADA2 protein (p.Met436Thr). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant is present in population databases (rs775742013, gnomAD 0.002%). This missense change has been observed in individual(s) with polyarteritis nodosa (Invitae). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. ClinVar contains an entry for this variant (Variation ID: 1399986).
